Ali Amin Gandapur joined PTI and won the elections from Dera Ismail Khan in 2013. He served as Minister of KPK Revenue and Estate.

In 2018, he won the election from DI Khan again. He became a member of the Federal Cabinet of Imran Khan. He was appointed as Minister of Kashmir Affairs and Gilgit Baltistan.
